Somatic embryo derived multiple shoot regeneration studies in Bacopa monnierae

Abstract


Bacopa monnieri L. commonly known as “Brahmi” is a renowned medicinal plant which belongs to the family Scrophulariaceae. It possesses anti-inflammatory, analgesic and antipyretic activity. Effective in vitro regeneration of brahmi has been achieved via young leaf derived somatic embryo cultures. The leaf explants were inoculated on MS medium with 2, 4- D (2, 4-D 0.5 mg/l) for callus induction. These calli buds were sub cultured on MS medium supplemented with various concentrations of 2, 4-D (0.75-1.75mg/l) along with BA (0.5-2.0mg/l) for somatic embryogenesis. Somatic embryogenesis was effective on MS medium with 2, 4-D 0.75mg/lt within three weeks of time. The initiated shoot buds were transferred onto (MS) medium with varied concentrations of BA (0.5-1.5mg/lt) for induction of multiple shoots. High percentage 99%of shoot induction were observed on media containing BA 0.5mg/lt++ IBA 0.3mg/lt. Efficient rooting 95% was noted on MS media with IBA 1.0mg/lt. In vitro regenerated plantlets were further acclimatized in the field.
